The second seeded Kyrgios spent one hour and 47 minutes in his first career win over Isner and snap the hard-serving American's 15-match winning streak here on Sunday, reports Xinhua.
"Returning is very crucial against John," said Kyrgios. "That serve is world class."
Kyrgios will rise to a career high ranking of 16 with this win.
He lost to Isner at Association of Tennis Professionals (ATP) Tour tournaments in Madrid and Montreal last year.
