Karkare worked for the National Productivity Council and Hindustan Lever before joining Indian Police Service. Since 1985, Karkare was posted at Nanded, Akola, Thane and Bhiwandi and in Mumbai as a deputy commissioner of police in the economic offense wing. Karkare also worked for sometime with the research and analysis wing – the country's external intelligence agency. Before handling the post of Anti Terrorist Cheif, Karkare was handling the post of Joint Commisioner of police (Administration).

Recent sensetive cases handled by his team include the Mumbai July 2006 local train blasts and the Sepember 29 Malegaon blast.
