What's the best way to end a relationship when it doesn't seem like anything is going to happen?
I don't know what you mean by things not happening but, when faced with stagnation, it's best to be open about it and choose to fix it or move on. The best way to end something is the same as the way to begin: With honesty and transparency, because both people in a relationship owe that to themselves.

My sister has always been pampered, ever since we were little. She has always taken the toys she wants, the clothes she prefers and even the friends she likes. I was okay with this because I am not as bossy as she is and my parents have never differentiated between us in any way. The problem is my boyfriend, who my sister likes a lot as well. He doesn't like her, and we have been together for a few months, but she always tries to make me look bad in front of him in the hope that he dumps me and chooses her. I have no proof of this but I have known her all my life and recognise why she does some things. I don't want to get into a fight with her, but I don't know how to deal with this either. I want to put my foot down and ask her to stop interfering with my life. What should I do?
Sibling rivalry can be intense, but this does sound rather dramatic. Your boyfriend chooses to be with you though, not your sister. You don't have to worry about looking bad in front of him if he understands that your sister behaves the way she does because she is insecure about a few things. You don't have to fight, but you can always draw lines about where members of your family can and cannot intrude. If you don't like what she's doing, what's stopping you from telling her to back off?
