The number one seed proved he is still a force to be reckoned with on clay as he blasted Carreno Busta away in 31 minutes in the first set.
An hour's rain break disrupted Nadal's momentum and he was broken twice in the second set, but still did enough to get through in straight sets.
Number two seed and reigning champion David Ferrer also made serene progress as he beat Nicolas Jarry in straight sets, recording a 6-3, 7-6 victory.
