Costa, who has scored 16 goals for the Blues this season, has been dogged by hamstring injuries and has not been named in Vicente del Bosque's group.
Man United midfielder Juan Mata, who has 41 caps is also omitted from the squad.
Former Chelsea and Liverpool striker Fernando Torres has also been unable to force his way into the squad, despite an impressive season with Atletico Madrid.
Torres has scored 10 goals in the last 19 games, helping Atletico to the Champions League final, but that has not been enough to earn a place.
Arsenal midfielder Santi Cazorla is another notable absentee although the uncapped pair Lucas Vazquez and Saul Niguez, of Real Madrid and Atletico Madrid respectively
are included.
